Transgenic / Knockout Animal Program

Overview of Services

The TKAP has been in operation at COH since the early 1990s, and is focussed on the production of genetically-modified animal models for basic and translational research. The TKAP produces transgenic mice by pronuclear microinjection, or by sub-zonal injection of pseudotyped self-inactivating lentiviral vectors. The TKAP generates mouse models carrying targeted gene inactivation (knock-out), or specific modifications of the nucleotide sequence of genes (knock-in) using mouse embronic stem cells (mESC). The TKAP also performs genome engineering with sequence-specific nucleases (TALENs & CRISPR/Cas9) in mESC, or directly in mouse zygotes. In addition the TKAP cryopreserves mouse embryos and gametes, and rederives mouse strains by in vitro fertilization and embryo transfer.
