Lipidomics Core

Objectives

The principal objective of the Lipidomics Core is to provide expertise and facilities to generate highly specific and ultra sensitive measurements of lipid mediators for funded and proposed research by investigators at Wayne State University and its partnering institutions. Current focus of the Core is on the analysis of fatty acyl lipids, sphingolipids, phospholipids, and sterol lipids. These include inflammatory mediators such as eicosanoids (e.g. prostaglandins, thromboxanes, leukotrienes, hydroxy fatty acids, etc.), resolution mediators such as lipoxins and resolvins, sphingosine-1-phosphate, ceramides, phosphatidylcholines, phosphatidylethanolamines, phosphatidylserines, inositols, and steroids. The biomarkers to be measured also include physiologically active oxidative stress markers (e.g. isoprostanes). Such assays are critical for assessing specific hypotheses regarding basic mechanisms of inflammation, carcinogenesis and other diseases, the roles of causal agents and covariates (e.g., nutrition), bioactivation and detoxification, DNA repair and biomarkers of exposure in occupational and environmental epidemiology. By providing this technology, the Core will provide members of the research community a major opportunity for groundbreaking studies.

Overview of Services

The Core will provide necessary equipment and expertise to enable researchers to identify and/or measure lipid biomarkers with state-of-the-art LC-MS/MS techniques. Methods are in place to comprehensively analyze the bioactive active lipids that are involved in inflammation and cancer (prostaglandins, thromboxane, HETEs, resolvin, protectins, etc.).

Samples for analysis can be liquids (tissue culture supernatants, tissue homogenates or biological fluids such as plasma, urine, etc.) or solid tissues. The samples can be processed and prepared for LC-MS analysis by the Core or by the investigators (with appropriate training from the core) following Standard Operating Procedures developed by the Core. Appropriate analytical targets are set in consultation with the investigator and methods will be developed, if necessary. As mentioned above, analytical methods for the analyses of the entire range of eicosanoids and docosanoids are already established by the core.

Analytical data will be emailed back to the researcher in a spreadsheet form.

All Core services start with an in-depth consultation with the researcher, free of cost, and a detailed quote will be provided for the agreed services and must be approved by the users before samples can be shipped and analyses begin.
